ABSTRACT

Objective To analyze the protective effect of tanshinone Ⅱ A sodium sulfonate on rat skeletal muscle after exhaustive exercise in high temperature environment.Methods Ninety rats were divided into the observation group,blank group and control group,30 cases in each group.The rats in the blank group received no intervention,while which in the observation group and control group received the exhaustive exercise intervention under high temperature environment.The rate in the control group was injected with 8 mg/kg normal saline by femoral vein at 5 h after exhaustive exercise under high temperature environment,while which in the observation group was injected with 40 mg/kg tanshinone Ⅱ A sodium sulfonate by femoral vein.The arterial blood in each group was collected by tail vein before killing the rat,and the right leg gastroc nemius muscle of each group was removed and frozen.The serum CK,LDH activity,TNF-alpha,IL-6,IL-1β,MDA,SOD and GSH-PX levels in skeletal muscles were examined.The rat skeletal muscle samples were conducted the HE staining for observing the morphological change.Results The levels of serum TNF-α,IL-6 and IL-1β and activity of CK and LDH in the observation group were significantly lower than those in the control group,while higher than those in the blank group,the difference was statistically significant (P<0.05).The skeletal muscle MDA,SOD and GSH-PX levels had statistical difference between the observation group and other two groups (P<0.05).The skeletal muscle structure was disordered,and the distribution of muscle nuclei was uneven.The structure of rat skeletal muscle in the blank group was normal,and the muscle nuclei were evenly distributed.The morphology and structure of skeletal muscle in the observation group were normal,but the distribution of muscle nuclei was slightly uneven.Conclusion Tanshinone Ⅱ A sodium sulfonate can effectively protect skeletalmuscle after exhaustive exercise inhigh temperature environment.

ABSTRACT

Objective To observe the clnical effects of influence of tanshinone type ⅡA sulfonate on preventing hepatic artery thrombosis after transplantation.Methods A total of 60 patients after liver transplantation were randomly individed into the treatment group and control group, each 30 patients. The treatment group received tanshinone ⅡA sodium sulfonate treatment (60 mg qd, ivgtt continuous 10d) , while the control group used conventional heparinization. The blood coagulation index and the thrombelastograph variables were detected after 7 days and the hepatic artery resistance index (RI) was detected by using Doppler ultrasonography. The postoperative complications and mortality rates were analyzed.Results Although it had little improvement on the coagulation function after liver transplantation, tanshinone ⅡA sodium sulfonate had significant improvement on the time of thrombelastograph parameters reaction (6.35 ± 1.59 minvs. 5.21 ± 1.37 min,t=2.453) and maximum amplitude (58.07 ± 5.42 mmvs. 61.67 ± 5.63 mm,t=-2.532). It showed that RI have significantly statistical difference between the two groups after treatment (0.73 ± 0.11vs. 0.62 ± 0.10;t=-2.948,P<0.01). During the trial, the control group had 2 cases of postoperative complications, HAT and bleeding.Conclusions The Tanshinone ⅡA sodium after liver transplantation can improve the clotting mechanism, preventing HAT.

ABSTRACT

AIM To prepare the liposomes loaded with celastrol,tanshinone Ⅱ A and tanshinone Ⅱ A sodium sulfonate and to evaluate the effects of preparation technologies and entrapment materials on the encapsulation efficiencies.METHODS The liposomes for three constituents were prepared by film dispersion method and reverse phase evaporation method,respectively.50% Lecithin high potency,80% egg yolk lecithin,92% lecithin high potency and complex phospholipid (5 ∶ 1 for ratio of dipalmitoyl phosphatidylcholine to 50% lecithin high potency) were served as entrapment materials.The encapsulation efficiencies were thus calculated based on the measured ean sizes and polydispersity of the liposomes.RESULTS Celastrol and tanshinone Ⅱ A sodium sulfonate liposomes accomplished by reverse phase evaporation method got the highest encapsulation efficiency when entrapped with 80% egg yolk lecithin and 92% lecithin high potency,respectively,which were higher than those prepared by film dispersion method.And celastrol liposomes exhibited an even superior performance than tanshinone Ⅱ A sodium sulfonate liposomes.Tanshinone Ⅱ A lipisome prepared by film dispersion method and then entrapped with 50% lecithin high potency obtained its the highest value of encapsulation efficiency,which was much lower than those of another two liposomes.CONCLUSION Reverse phase evaporation method is appropriate for hydrophobic celastrol with multiple hydrogen bonds and hydrophilic sodium tanshinone Ⅱ A without hydrogen bonds to prepare their liposomes,while film dispersion method is applicable to hydrophobic tanshinone Ⅱ A without hydrogen bonds.

ABSTRACT

OBJECTIVE:To investigate the effects of TanshinoneⅡA sodium sulfonate injection on levels of P-selectin,glial fi-brillary acidic protein (GFAP),vascular endothelial growth factor (VEGF) and neurological function in patients with acute cere-bral infarction. METHODS:A total of 114 patients with acute cerebral infarction selected from Lianyungang First People's Hospi-tal during Apr. 2013-Apr. 2016 were divided into control group and observation group according to random number table,with 57 cases in each group. Control group was given routine treatment. Observation group was additionally given Tanshinone ⅡA sodium sulfonate injection 40 mg 0.9% sodium chlonride injection 250 mL,ivgtt,qd. A treatment course lasted for 7 d,and both received 2 courses of treatment. NIHSS scores,the levels of serum P-selectin,GFAP and VEGF were compared between 2 groups before treatment and after 7,14 d of treatment. The occurrence of ADR was also compared. RESULTS:Before treatment,there was no statistical significance in above indexes between 2 groups(P>0.05). Compared to before treatment,NIHSS score,the levels of se-rum P-selectin and GFAP in 2 groups were decreased significantly after 7,14 d of treatment,while the serum level of VEGF was increased significantly. These indexes of 2 groups after 14 d of treatment were significantly better than 7 d of treatment,except for NIHSS score. Above indexes of observation group was significantly better than those of control group during corresponding period, with statistical significance (P<0.05). No obvious ADR was found in 2 groups. CONCLUSIONS:For acute cerebral infarction, Tanshinone ⅡA sodium sulfonate injection can significantly reduce the levels of serum P-selectin and GFAP,improve VEGF level and promote the recovery of neurological damage with good safety.

ABSTRACT

Objective To observe the effects of Tanshinone Ⅱ A sodium sulfonate (TSS ) on ischemia/reperfusion (I /R) induced cardiac injury in male (Sprague-Dawley,SD ) and explore its mechanisms.Methods Rats were subjected to a 30 min coronary arterial occlusion followed by 24 hours reperfusion.The survival rats were randomly (random number)divided into sham group (Sham group,n =10),ischemia reperfusion group (I /R group,n =10),low dose of TSS group (TSS-L group,n =10), medium dose of TSS group (TSS-Mgroup,n =9),high dose of TSS group (TSS-H group,n =9).A MAP heart function analysis system was used to measure hemodynamic variables,and TTC staining method was used to detect the myocardial infarct size.The levels of Bcl-2,Bax,Caspase-3,Lc3B/Lc3A,Beclin-1 and high mobility group box1 (HMGB1)were detected by western blot method.All data were analyzed by using One-way analysis of variance (ANOVA)(LSD-t test).Results Cardiac function in I /R group was lower than that in Sham group,and that was significantly improved by pretreated with TSS (P 0.05 ).The percentage of myocardial infarct size in TSS pretreatment group was significantly smaller than that in I /R group (P <0.05 ).Compared with Sham group,levels of Caspase-3 and Bax increased,and the Bcl-2 content was reduced obviously in I /R group (P <0.05).TSS pretreatment significantly down-regulated the levels of Caspase-3 and Bax protein (P <0.01).At the same time,the level of Bcl-2 was increased in all TSS pretreatment groups (P <0.01).Compared with Sham group,the ratio of Bcl-2 /Bax in I /R group was lower (P <0.05),and that was elevated in TSS groups (P <0.05 ).The change of autophagy related protein beclin-1 and Lc3B/Lc3A was in similar trend,and the levels of beclin-1 and Lc3B/Lc3A in I /R group were lower than that in Sham group (P <0.05),and those were raised in TSS pretreatment groups (P<0.05).The level of HMGB1 in I /R group was higher than that in Sham group (P <0.05),and compared with I /R group,the level of HMGB1 significantly decreased in TSS pretreatment groups (P <0.01 ). Conclusions The tanshinone ⅡA sodium sulfonate can protect the myocardium from ischemia/reperfusion injury and the mechanism may be attributed to the inhibition of cell apoptosis and activation of cell autophagy.
